About Melanotan-1

Melanotan-1, also called afamelanotide, is a 13 amino acid analogue of alpha-melanocyte stimulating hormone. It differs from the natural hormone at two positions, which is enough to make it far more resistant to enzymatic breakdown while leaving its receptor behaviour largely intact.
The critical difference from MT2 is selectivity. Melanotan-1 is a comparatively selective MC1R agonist, and MC1R is the receptor on melanocytes that governs eumelanin production. MT2 is a broader agonist that also hits MC3R and MC4R, which is where its libido effects, its nausea and its appetite suppression come from. Melanotan-1 largely does not touch those receptors, so it produces pigmentation with markedly fewer of the side effects MT2 is known for.
It is a licensed medicine in Europe and the United States under the brand name Scenesse, an implant used to prevent phototoxic reactions in erythropoietic protoporphyria. That approval is for a rare photosensitivity disorder, not for cosmetic tanning, and the implant is a different product from the material supplied here.

Where the evidence sits: afamelanotide has completed human trials and holds a European and US licence for one rare disease, so its pharmacology and its pigmentation effect are established. That licence does not extend to cosmetic use, and the long term consequences of driving melanogenesis in people without that disorder have not been characterised. Existing moles darkening is a documented effect.

The pen format
Melanotan-1 has a shorter half life than MT2 and the loading protocols in the literature are daily, so a 300 unit pen with 50mcg steps is a considerably better fit than reconstituting a vial every few days.

Choosing between Melanotan-1, MT2 and the nasal sprayThis decision is almost entirely about side effects. MT2 is cheaper, more potent for pigmentation, and hits MC3R and MC4R as well as MC1R, which is why nausea, flushing, appetite suppression and spontaneous erections show up so consistently in MT2 reports. Melanotan-1 is the selective one: slower and more expensive per tan, with far less of that.If the extra receptor activity is unwanted, Melanotan-1 is the reason to pay more. If it is wanted, or tolerated, MT2 does the job for less.
